Description

In this contribution, mathematical models describing the interaction between leukemic cells and CAR-T cells are developed and analyzed from the perspective of mathematical oncology. As a starting point, we consider the system of nonlinear ordinary differential equations proposed by \cite{PerezGarcia2021CART}, which describes the joint dynamics of CAR-T and tumor cells by incorporating proliferation, exhaustion, and fratricide effects. Building on this framework, the model is extended by introducing an additional term representing CAR-T cell reinjection, allowing for the analysis
of both continuous constant administration and impulsive reinjection schemes. The analysis focuses on the qualitative behavior of the system, including equilibrium stability, the occurrence of bifurcations, and the dynamic response under different therapeutic regimes. In particular,
the study of the continuous reinjection case shows that, for suitable values of the parameter u,the system can stabilize at a state characterized by a low but persistent CAR-T cell population capable of eliminating small tumor reappearances and preventing relapse. For other parameter ranges, the model predicts the coexistence of CAR-T and leukemic cells, describing a scenario of chronic disease control rather than complete eradication, suggesting that continuous dosing may play a key role in the long-term maintenance of tumor control.
